外源性分化因子对鸡前脂肪细胞分化之影响

外源性分化因子對雞前脂肪細胞分化之影響

摘 要
本試驗比較在不同外源性分化因子誘發下,雞隻脂肪前身細胞的分化表現,以期建立適合的雞隻脂肪細胞分化培養模式。將取自雞隻腹脂的脂肪前身細胞培養於胎牛血清(FBS)、雞血清(CKS)、雞血清外並添加胰島素與dexamethasone(CID)、或是雞血清外並添加胰島素、dexamethasone與3-methyl-1-isobutylxanthine(CIDM)等四種培養模式誘發細胞分化。在分化過程中以顯微鏡觀察脂肪細胞型態變化,並於分化後第六天,分析脂肪細胞三酸甘油酯堆積情形與GPDH活性,另外以相對定量反轉錄聚合酶連鎖反應分析分化相關基因表現。實驗結果顯示與胎牛血清培養模式相較,三種含雞血清培養模式皆可顯著促進分化細胞三酸甘油酯堆積、GPDH活性以及PPAR? 與FAS mRNA表現,然而以上分析結果在三種含雞血清培養模式間並無顯著差異。本試驗結果顯示,雞隻脂肪前身細胞在含有雞血清的培養條件下,即可成功地促進脂肪細胞分化;而在含雞血清的培養液中,加入胰島素、dexamethasone與3-methyl-1-isobutylxanthine對脂肪細胞分化無顯著之影響。

Effects of Exogenous Inducers on Chicken Preadipocyte Differentiation

Abstract
Effects of different combinations of differentiation inducers on chicken preadipocyte differentiation were studied to aid the establishment of optimal culture model for chicken aidpocyte differentiation. Preadipocytes collected from chicken abdominal adipose tissues were cultured in media containing fetal bovine serum (FBS), chicken serum (CKS), chicken serum plus insulin and dexamethasone (CID), or chicken serum plus insulin, dexamethasone, and 3-methyl-1-isobutylxanthine (CIDM), to induce differentiation. The morphology of preadipocytes during the process of differentiation was monitored. Triacylglyceride content and GPDH activity assays were also performed using adipocytes 6 days after the induction of differentiation. Additionally, mRNA levels of differentiation related genes, i.e. PPARγ and FAS, were assayed by RT-PCR analysis. The results of these assays revealed that all of three culture models with CKS effectively induced differentiation of preadipocyte comparing with FBS model, however, no significant differenc

es in the induction capacity were observed among these three culture models. These results suggest that chicken serum is capable of inducing chicken preadipocyte differentiation and the exogenous inducers, insulin, dexamethasone, and 3-methyl-1-isobutylxanthine, do not facilitate preadipocyte differentiation in the presence of chicken serum.
